การตรวจ RT-PCR (Polymerase chain reaction) เป็นการ Swab เก็บตัวอย่างเชื้อบริเวณลำคอ และหลังโพรงจมูก (เช่นเดียวกันกับตรวจ Antigen) แต่จะทราบผลใน 2-3 วัน เนื่องจากต้องมีการวัดผลผ่านห้องปฏิบัติการ ถือเป็นการตรวจที่มีความถูกต้องแม่นยำมากกว่า แต่จะใช้เวลาในการวินิจฉัยตัวอย่างเชื้อนานกว่า และเป็นการตรวจที่แนะนำจาก WHO เนื่องจากสามารถตรวจหาเชื้อในปริมาณน้อยไ
